The IDK® S100A12 ELISA is designed for the quantitative determination of S100A12.
A monoclonal antibody specific for S100A12 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells, and the immobilized antibody binds any S100A12 present. After washing away any unbound substances, an HRP-conjugated polyclonal antibody specific to S100A12 is added to the wells. Following a wash to remove any unbound antibody HRP-conjugate, the remaining conjugate reacts with the HRP substrate tetramethylbenzidine.
Adding an acidic solution stops the reaction, and the absorbance of the resulting yellow product is measured at 450 nm. The absorbance is proportional to the concentration of S100A12.
A dose-response curve of the absorbance unit (optical density, OD at 450 nm) vs. concentration is generated using the values obtained from the standard. The presence of S100A12 in the samples is determined directly from this curve.
