Test Principle of the Human Anti-thyroglobulin Antibody CLIA Kit

The Human Anti-Thyroglobulin Antibody (Anti-Tg) CLIA Kit is designed, developed, and produced for the quantitative measurement of human Anti-Tg level in serum samples. The assay utilizes a two-site “sandwich” technique with two antigens that bind to different epitopes of Anti-Tg.

The sample, biotin-labeled Tg antigen, another Tg antigen labeled with acridinium ester and streptomycin-coated magnetic micro beads are mixed thoroughly and incubated, forming an antigen-antibody sandwich complex, which is then bound to the magnetic micro beads through the interaction between biotin and streptavidin.

After precipitation in a magnetic field, the supernatant is decanted, and then a wash cycle is performed to remove any remaining substances that are not bound to the magnetic micro beads.

Subsequently, the washed compound is sent into the measurement chamber where trigger solution is automatically added to initiate a chemiluminescence reaction. The light signal is measured by a photomultiplier as relative light units (RLUs), which is proportional to the concentration of Anti-Tg present in the sample.

The test result is automatically calculated by the system according to the working curve.
